GPIO/SPI to Microcontroller

Hi guys, im looking to create a mouse controller on my SBC, i want to use a mouse via USB input as usual on the SBC, then read mouse signal/position and output it though a microcontroller to go into my host PC maybe either GPIO or SPI is this possible?
